Intact mass of a heterogeneous therapeutic protein was determined by Direct Mass Technology (Orbitrap based charge detection mass spectrometry), proving mono PEGylation of the biologic. Furthermore, using multiple enzyme combinations, major PEGylation sites of the protein were mapped by finetuning instrument parameters and chromatographic conditions.
