Atlantic herring oocyte and sperm proteomes

Description

As part of a major genomic study of the evolution of Atlantic herring (Clupea harengus), deep proteomes were obtained from the oocytes and sperm of herring. Three biological replicates of each tissue were used. The proteins were extracted and fractionated using basic reversed-phase (bRP) chromatography. Twelve fractions from each replicate were analyzed using LC-MS/MS. Mass spectrometry data were acquired in data-dependent acquisition (DDA) mode using an Orbitrap Exploris 480 instrument. The raw data were processed and searched using MaxQuant. Protein identification was performed using a reference proteome database of Clupea harengus supplemented with sequences of three additional LRRC8C2 alleles. LRRC8C2 is a sperm-specific volume-regulated ion channel that is the focus of a major evolutionary study. As a result, more than 4,000 and 6,000 protein groups were identified in sperm and oocytes, respectively. The Baltic North allele of LRRC8C2 was unambiguously expressed.
