The goal of this experiment is to identify changes in TEAD4 protein interactions upon treatment with TEAD small molecule inhibitors. NCI-H226 cells were treated for 24h with DMSO vehicle control or a TEAD inhibitor, GNE-7883 or Compound 2. Immunoprecipitation of TEAD4 was performed and the resulting protein samples were processed for tandem mass tag (TMT) labeling and quantitative proteomic analysis. Pooled TMT-labeled samples were fractionated into six fractions and the experiment was performed in triplicate.
