Ziziphus jujuba metabolomics - cyclopeptide alkaloid biosynthesis

0.3g of fresh weight Ziziphus jujuba leaves were transferred to an MP Biomedicals Tissuelyzer tube (2mL, Cat: 5076200, 5068002) with 10-15 2.3mm Zirconia silica beads (Cat: NC0419764). 0.3g of fresh weight Ziziphus jujuba stem were transferred to a metal homogenizer tube with a 0.25in. steel ball (MP Biomedicals, MP116991006). Sample tubes were frozen at -80C for at least 30min, and ground in a MP Biomedicals FastPrep-24-5G Tissuelyzer for 60s at 6m/s. 1mL of 80% methanol was added to the ground plant material in the tissuelyzer tube and ground another 20s in the Tissuelyzer for 20s at 6m/s. The tubes were then incubated at 60C for 10min in a waterbath, centrifugated at 16000g at room temperature for 5min, and the supernatant was filtered with Cytvia syringeless LCMS filters (UN503NPEORG). The plant methanolic extracts were then transferred to LCMS vials analyzed by LC-MSMS with the following parameters: LC - Phenomenex Kinetex 2.6um C18 reverse phase 100A 150x3mm LC column, LC gradient: solvent A = 0.1% formic acid; solvent B = acetonitrile (0.1% formic acid), 0min 10%B, 5min 60%B, 5.1min 95%B, 6min 95%B, 6.1min 10%B, 9.9min 10%B, 0.5mL/min, 30C, MS: positive ion mode, full MS, resolution 70000; mass range 400-1200m/z, ddMS2 (data-dependent MS/MS), resolution 17,500; AGC target 10e5, loop count 5, isolation width 1.0m/z, collision energy 25eV and dynamic exclusion 0.5s.
