Description
Inter-tumor heterogeneity is a reflective of the coalescence of a myriad of genomic, transcriptional, translational, and post-translational molecular features. To investigate the contribution of protein glycosylation in tumor heterogeneity in high-grade serous ovarian carcinoma (HGSC), we performed mass-spectrometry (MS)-based glycoproteomic characterization of 119 TCGA HGSC tissues. These HGSC tumor tissues have been previously characterized at the genomic and transcriptomic levels, as well as the proteomic and phosphoproteomic levels, by The Cancer Genomic Atlas (TCGA) consortia and the Clinical Proteomic Tumor Analysis Consortium (CPTAC), respectively. Employing two glycoproteomic strategies, Solid-Phase Extraction of Glycosite-containing peptides (SPEG) for glycosite analysis, and intact glycopeptides for investigation of glycosite-specific glycans (IGPs), we profiled the N-linked glycoproteome in HGSCs, identifying and quantifying glycosites and attached glycan structures.
