The precleared whole cell P. aeruginosa PA14 lysates underwent an overnight incubation with bead-immobilized N-104 peptide (or C-95 as a negative control peptide) under physiological conditions. Subsequent steps included washes and a gradual step-gradient elution ranging from 50 to 500 mM KCl. The eluate obtained at 500 mM KCl was then analyzed using tandem mass spectrometry to identify PA14 proteins that bind to N-104.
