For SHLP2_ carboxyLinkTM kit set, we used carboxyLinkTM kit to create a SHLP2 peptide column and a scrambled peptide column as a control to identify direct SHLP2 binding proteins. The mitochondria protein extract were used to detect direct binding partners of SHLP2.
For SHLP2_APEX set, APEX2 proximity labeling method is used to identify the binding proteins of SHLP2, a mitochondrial microprotein. Plasmid harboring SHLP2-APEX fusion proteins (SHLP2-APEX-myc) were transfected into cells The biotinylated proteins were enriched by streptavidin beads pulldown. Two controls were used the APEX control (Flag-APEX-myc) and MA control (MTS (mitochondrial targeting sequence)-APEX-myc).
