From yeast cells expressing Flag-Hst2, soluble (cytosolic) fractions were collected. Cells expressing untagged Hst2 served as control. IP was performed on both experimantal and conrol samples with Anti-Flag agarose beads. Flag-Hst2 and bound proteins were precipitated und purified running a short SDS-gel. LFQ was used to identify binding partners and search for S, T, and Y phosphorylation were performed separately.
