MGRN1, a membrane-anchored E3 ligase, uses multiple transmembrane adapters-including newly identified ATRN and ATRNL1-to target specific surface receptors for ubiquitination and degradation. These adapters bind the RING domain of MGRN1 and enable it to regulate melanocortin receptors MC1R and MC4R in a manner similar to its known control of Smoothened. Loss of MGRN1 increases surface levels of these receptors and boosts eumelanin production, revealing an expanded and unifying mechanism for receptor-specific regulation by membrane-tethered E3 ligases.
