Engineered bacterial strains with bile salt hydrolase that changed expression levels with a high-fat diet and diurnal changes were grafted into mice. The fecal samples from these mice were collected and extracted with 50% methanol for LC-MS/MS metabolomics. The sample extracts were chromatographed using a Phenomenex polar C18 column, and MS/MS data was acquired on a Thermo Q Exactive in positive ionization mode.
