Plants were treated with a cPEP for luciferase (PEP Luc) or a scrambled version of this peptide (4 biological replicates each). Leaves were snap frozen in liquid nitrogen and grounded into powder. The powder was resuspended in tris 50 mM and 5% SDS, sonicated and centrifuged at 14,000 g for 10 min. A BCA protein assay was performed to estimate protein concentration in each sample. 50 ug of proteins were then loaded on a 12% Tris-glycine gel. Proteins were concentrated into one band and in gel digestion was performed as previously described (PMID: 25561571). 500 ng were then injected on a Thermofisher Exploris 480.
