N-glycoproteomic and O-man glycoproteomic analysis of N-cadherin derived from wild-type(WT) and POMGnT1-deficient HEK293T cells. Proteolytic digestion was performed with trypsin or proteinase K. Glycopeptide enrichment was achieved using cotton-HILIC SPE. Glycopeptides were fragmented by HCD.step and HCD.low. For O-man glycoproteomics N-glycan release by PNGase F was included.
