The analysis of high-abundance megadalton protein complexes in Dictyostelium discoideum cell lysates was conducted as follows: the solubilized protein fraction was subjected to size-exclusion chromatography isolating fractions corresponding to the megadalton region, followed by ion-exchange chromatography to further resolve the protein complexes. The fractions obtained after both chromatographic steps were then analyzed using mass spectrometry to identify the constituent protein complexes, which provided the necessary data for cryo-EM.
