The inner and outer membranes were isolated from three biological replicates of two different strains of Francisella tularensis (LVS and Schu S4). Peptides were generated by proteolytic digestion with trypsin. Samples were analyzed on a quadrupole time-of-flight TripleTOF 5600 mass spectrometer and the data searched using Protein Pilot. Weighted spectral counting was performed using peptides that were at least 99% confident. Additionally, the membrane preparations were used to inoculate mice against a challenge with either LVS or SchuS4 strains. Immunoreactive bands from the mouse vaccination study were also extracted, digested with trypsin, and the spectral counts calculated.
