To elucidate the specific proteins involved in bone formation that are modified by 4-HNE, we performed LC-MS-based proteomics analysis. This approach identified several candidate proteins, including MAP kinase-activated protein kinase 3 (MAPK3), plastin-3 (PLS3), and integrin-linked protein kinase (ILK), all of which are implicated in bone formation or osteoblast differentiation. Notably, 4-HNE-modified ILK was significantly elevated in the GPX4-inhibited group (RSL3 treatment) compared to the control group.
